Draft Prokaryotic two - component systems and the OmpR / PhoB superfamily

چکیده
23 In bacteria, two-component regulatory systems (TCSs) are the critical information-processing 24 pathways that link stimuli to specific adaptive responses. Signals perceived by membrane 25 sensors, which are generally histidine kinases (HKs), are transmitted by response regulators 26 (RRs) to allow cells to cope rapidly and effectively with environmental challenges. Over the 27 past few decades, genes encoding components of TCSs and their responsive proteins have 28 been identified, crystal structures have been described, and signaling mechanisms have been 29 elucidated. Here, we review recent findings and interesting breakthroughs in bacterial TCS 30 research. Furthermore, we discuss structural features, mechanisms of activation/regulation, 31 and cross-regulation of RRs with a focus on the largest RR family, OmpR/PhoB, to provide a 32 comprehensive overview of these critically important signaling molecules. 33

Structure of the response regulator VicR DNA-binding domain
The response regulator VicR from the Gram-positive bacterium Enterococcus faecalis forms part of the two-component signal transduction system of the YycFG subfamily. The structure of the DNA-binding domain of VicR, VicR(c), has been solved and belongs to the winged helix-turn-helix family. It is very similar to the DNA-binding domains of Escherichia coli PhoB and OmpR, despite low sequence simi...

Two-component signal transduction systems in bacteria are a primary mechanism for responding to environmental stimuli and adjusting gene expression accordingly. Generally in these systems a sensor kinase phosphorylates a response regulator that regulates transcription. Response regulators contain two domains: a receiver domain and an effector domain. The receiver domain is typically phosphoryla...
متن کاملSystem-level mapping of Escherichia coli response regulator dimerization with FRET hybrids
SUMMARY Two-component signal transduction, featuring highly conserved histidine kinases (HKs) and response regulators (RRs), is one of the most prevalent signalling schemes in prokaryotes. RRs function as phosphorylation-activated switches to mediate diverse output responses, mostly via transcription regulation. As bacterial genomes typically encode multiple two-component proteins for distinct ...
متن کاملAmino acids important for DNA recognition by the response regulator OmpR.
Response regulators undergo regulated phosphorylation and dephosphorylation at conserved aspartic acid residues in bacterial signal transduction systems. OmpR is a winged helix-turnhelix DNA-binding protein that functions as a global regulator in bacteria and is also important in pathogenesis. A detailed mechanistic picture of how OmpR binds to DNA and activates transcription is lacking. We use...
متن کاملInterdomain linkers of homologous response regulators determine their mechanism of action.
OmpR and PhoB are response regulators that contain an N-terminal phosphorylation domain and a C-terminal DNA binding effector domain connected by a flexible interdomain linker. Phosphorylation of the N terminus results in an increase in affinity for specific DNA and the subsequent regulation of gene expression. Despite their sequence and structural similarity, OmpR and PhoB employ different mec...
